Specimen Requirement
Cervix: Visualize the cervix using a speculum without lubricant. Remove mucus and secretions with a swab and
discard the swab. Firmly, yet gently, sample the endocervical canal with a Culturette swab. Place the swab in the transport tube.
Female urethra: Collect the specimen 1 hour after the patient has urinated. Remove exudate from the urethral
orifice. Collect discharge material on a Culturette swab by massaging the urethra against the pubic symphysis through the vagina. Place the swab in the transport tube.
Male urethra: Insert a Mini-Tip Culturette swab 2-4 cm into the urethral lumen, rotate the swab, and leave it in
place for 2 seconds to facilitate absorption. Place the swab in the transport tube.
Prostate: Cleanse the glans with soap and water. Massage the prostate through the rectum. Collect fluid on a
Culturette swab or in a sterile container. If collected on a swab, place the swab in the transport tube.
Note: Molecular testing is recommended for Neisseria gonorrhoeae, Chlamydia trachomatis, Trichomonas
vaginalis, Ureaplasma, Mycoplasma, Haemophilus ducreyi, HPV, and HSV. Serology is the test of choice for Treponema pallidum. Laboratory consultation is recommended for unusual requests.
Rejection Criteria
Reject if specimen container is unlabeled or leaking.
Reject for inappropriate storage or transport.
Storage and Transport
Transport the specimen to the Microbiology Lab at room temperature within 2 hours.
If transport is delayed, specimens may be stored at room temperature for up to 24 hours before testing.
